滋阴益气活血法对衰老糖尿病大鼠胰腺组织caspase-3表达的影响

摘　　要：目的观察滋阴益气活血法对衰老及衰老糖尿病大鼠胰腺组织凋亡相关基因caspase-3表达的影响。方法实验分为正常组、衰老模型组、衰老糖尿病模型组、石斛合剂组。除正常组外,大鼠腹腔注射D-半乳糖致衰老,继以高脂高糖加腹腔注射链脲佐菌素(STZ)造成衰老糖尿病模型。疗程结束后取血测血脂及游离脂肪酸水平,取胰腺采用RT-PCR法检测凋亡相关基因caspase-3 mRNA表达水平。结果与正常对照组相比,衰老模型组大鼠血脂有升高趋势。血FFA水平上升(P<0.05),衰老糖尿病模型大鼠血脂升高(TG、TC、LDLC升高(P<0.01),HDLC降低(P<0.01)。血FFA水平也上升(P<O.05),衰老及衰老糖尿病模型组大鼠胰腺组织caspase-3 mRNA表达均明显升高(P<0.01);与衰老糖尿病模型组相比,石斛合剂组TG、TC、LDLC、FFA等指标均改善(P<0.01或P<0.05),尤其FFA明显下降(P<0.01),胰腺组织Caspase-3mRNA表达明显降低(P<0.01)。结论衰老糖尿病模型大鼠呈现脂代谢异常,胰腺组织细胞凋亡增多;而滋阴益气活血法(石斛合剂)可改善模型大鼠脂代谢,抑制胰腺组织细胞的凋亡。Objective To study the influence of the method of nourishing - ＂ yin＂, tonifying - ＂ qi＂ and activating - blood on caspase -3 mRNA expression associated with apoptosis in pancreas of aged diabetic rats. Methods The aged diabetic rat model was induced by D - galactose, enrich - diet and streptozotoeiu the concentrations of blood lipid and FFA in serum were measured, and detect caspase - 3 mRNA was assayed by RT - PCR. Results Compared with the control group, the level of FFA in aged group （ P 〈 0.05 ） and the levels of blood lipid （ TG, TC, LDLC ） and FFA were elevated （ P 〈 0.05 ）, the content of HDLC decreased in diabetic aged group（ P 〈 0.05 ） , and caspase -3 mRNA expression in aged and aged diabetic groups were evidently heightened（ P 〈 0. 01 ） ; compared to the aged diabetic group ,the level of blood lipid in DC treated group was improved（ P 〈 0.0l - 0.05 ） , and easpase - 3 mRNA expression was also evidently declined （ P 〈 0.05 ）. Conclusion It demonstrated that there had lipid metabolism disorders and pancreatic cell apoptosis in the aged diabetic group, and DC can impove lipid metabolism of diabetic rats via inhibiting the pancreatic cell apoptosis.
