补阳还五汤颗粒剂的药效学和急性毒性

摘　　要：目的 :用药理学方法研究古方补阳还五汤颗粒剂的药效学及其急性毒性 ,为治疗缺血性中风提供理论依据。方法 :补阳还五汤颗粒剂对实验性大鼠脑缺血的作用 ,对大鼠心电、血压、颈动脉血流量的影响 ,对凝血时间及血栓的影响。结果 :该颗粒剂对脑缺血大鼠脑指数显著降低 (P <0 .0 5) ,使颈总动脉血流量增加 (P <0 .0 1 ) ,降低脑血管阻力 (P <0 .0 1 ) ,延长凝血时间及抑制血栓的形成 (P <0 .0 1 ) ;急性毒性试验表明 ,对小鼠ig最大耐受量大于 2 7 5g·kg- 1 ,iv最大耐受量大于 3g·kg- 1 。结论 :补阳还五汤颗粒剂可增加颈总动脉血流量 ,降低脑血管阻力 ,延长凝血时间及抑制血栓的形成 ,对治疗脑缺血诱发的中风有一定意义 。OBJECTIVE:To examine the pharmacodynamics and acute toxicity of BHTG (buyang huanwu tang granule) originated from a traditional Chinese medical formula BHT on treating the wind stroke caused by cerebral ischemia.METHODS:Cerebral ischemia of rats,electrocardiogram,pressure,blood flow carotid,coagulation time,thrombosis in rats.RESULTS:BHTG could significantly lower the cerebral index of cerebral ischemia rats,resistance of cerebral blood vessel,the blood flows carotid and the coagulation time was increased,the thrombosis was also inhibited by BHTG in rats.In addition,the maximal tolerance to BHTG in mice was larger than 27.5 g·kg -1 or 3 g·kg -1 ,respectively following ig or iv BHTG.CONCLUSION:BHTG is safe and effective.
