天津地区人群中LOX-1基因3'-UTR多态性与冠心病的关系

摘  要:目的:研究天津地区人群中凝集素样氧化低密度脂蛋白受体-1(LOX-1)3′-非编码区(3′-UTR)188位点基因多态性情况,探讨LOX-1基因多态性与冠心病(CAD)及血脂水平的关系。方法:冠心病组170例,均经冠脉造影(CAG)确诊;对照组140例,均经CAG排查。应用聚合酶链反应-限制片段长度多态性(PCR-RFLP)法对研究对象的LOX-1基因3′-UTR188位点多态性进行分析,并用自动生化分析仪检测血浆血脂水平。结果:冠心病组和对照组比较,LOX-1基因3′-UTR 188位点基因型频率和等位基因频率差异均无统计学意义(均P>0.05);在总研究对象中T等位基因携带者(CT+TT)与CC基因型者血三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、脂蛋白(a)[Lp(a)]水平差异均无统计学意义(均P>0.05),冠心病组与对照组中的CT+TT基因型者以上血脂水平差异亦无统计学意义(P>0.05)。结论:天津地区人群中LOX-1基因3′-UTR188位点多态性与冠心病的发病及血脂水平均无明显关系,其不能作为冠心病的预测因素。Abstract Objective: To investigate the polymorphism of 3'-UTR 188 site of lectin-like oxidized low density lipoprotein receptor-1 (LOX-I) gene and to explore the correlation between the gene polymorphism of LOX-1 and coronary artery disease (CAD)and lipid levels in the population of Tianjin. Methods: A total of 170 cases confirmed by coronary arteriograghy (CAG) were included in CAD group and 140 healthy subjects confirmed by CAG were included in control group. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was used to analyze the polymorphism of 3'-UTR 188 site of LOX-1 gene and to compare its relationship with coronary artery disease between the two groups. The automatic biochemistry analyzer was used to detect plasma lipid levels of all subjects. Results: There was no significant difference in genotype frequency and allele frequency of 3'-UTR 188 site of LOX-1 between CAD group and control group (P 〉 0.05). There were no significant differences in triglyceride (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C) and lipoprotein (a) levels between T allele (CT+TI') and CC genotype (P 〉 0.05). There was no significant difference in lipid levels between CAD group and control group of subjects with CC+TT genotype. Conclusion: There is no significant correlation between the polymorphism of 3'- UTR 188 site of LOX-1, coronary artery disease and lipid levels in the population of Tianjin, therefore the polymorphism can not be used as a predictor of coronary artery disease.
