摘  要:目的:探讨了高效液相色谱法测定人血清中氨茶碱的药物浓度的方法。方法:采用symmetryshield RP18柱(5.0μm,3.9×150 mm)为色谱柱。流动相为甲醇∶水为15:85,流速1.0mL.min-1;Waters 2487双波长紫外检测器,检测波长272nm;乙酰苯胺为内标物。结果:线性范围为(4.4~33.0)μg.mL-1,r=0.999190,最低检测限0.16μg.mL-1;平均回收率为96.7%,高、中、低浓度样品的日间和日内测定相对标准偏差RSD均小于2%。结论:本法适用于氨茶碱的血药浓度测定。Objective: To establish a HPLC method for determining human serum level of aminophylline. Methods: Acetanilide was selected as the internal standard. A symmetryshield RP18 column(5μm,3.9×150mm)was used. The mobile phase, methanol-phosphate solution(15:85)was used at a flow rate 1.0 mL·min^-1. The detector is Waters 2487 Dual λ Absorbance Detector, and was set at the wavelength of 272 nm. Results: The calibration curve was linear in the range of 0.16 to 33.0μg·mL^-1. The average recovery was 96.7%; the RSD within-day and between-day were all less than 2%. Conclusion: This method is suitable for therapeutic drug monitoring of aminophylline.
