摘 要:目的:比较GSTp和GSTm对血管紧张素II诱导的血管平滑肌细胞过度增殖的抑制作用,以及对JAK/STAT3信号通路调控的影响,并探讨GSTp抑制作用的机制。方法:在VSMCs中过表达GSTp、GSTm及Y7F;MTT法检测细胞增殖,western blot检测STAT3磷酸化情况。结果:过表达GSTp和GSTm能够显著抑制血管平滑肌细胞的增殖及STAT3的磷酸化,GSTp的抑制效果较GSTm更佳,而Y7F抑制效果较差。结论:GSTp相比GSTm抑制血管平滑肌细胞增殖的效果更好,其作用机制可能是通过转移酶活性而下调ROS水平,从而抑制下游增殖相关信号通路。这证明GSTp在维持血管平滑肌细胞稳定性及动脉粥样硬化等心血管疾病中起重要的作用。Objective: To compare the inhibiting effect of VSMCs proliferation and STAT3 phosphoryla- tion by GSTp and GSTm, and study the mechanism of GSTp' s inhibiting effect. Method: VSMCs were transfected with GSTp, GSTm and Y7F plasmid and stimulated by Angiotensin 11. MTT assay was used to measure the effect of VSMCs proliferation, and western blot was used to measure phosphorylation of STAT3. Result: Both GSTp and GSTm inhibited VSMCs proliferation and STAT3 phosphorylation significantly, but GSTp had a stronger effects than GSTm; The inhibition by Y7F was weaker than GSTp and GSTm. Conclusion: GSTp inhibits proliferation of VSMCs is stronger than GSTm, and this function may connect with its transferase activity. These findings provide a new insight for understanding the role of GSTp in maintaining stability of vascular smooth muscle cells and decreasing the risk of atherosclerosis occurrence.
