补中益气汤对小鼠肺mBD-2表达的影响

摘　　要：目的探讨补中益气汤对小鼠肺天然免疫效应分子β防御素-2(mouseβ-defensin 2,mBD-2)的诱导作用。方法取C57BL/6小鼠,随机分为对照组和补中益气汤高、中、低剂量组,给予PBS和不同质量浓度的补中益气汤(2.0 g/mL、1.0 g/mL和0.5 g/mL)灌胃,7 d后取小鼠肺组织,分别用RT-PCR法和免疫组化法检测小鼠肺上皮细胞mBD-2 mRNA和蛋白表达情况。结果在基因水平和蛋白水平都发现对照组小鼠肺组织不表达mBD-2,而补中益气汤刺激后实验组小鼠肺上皮细胞表达mBD-2,且高剂量组作用效果强于低剂量组。结论补中益气汤可以诱导小鼠肺上皮细胞天然免疫效应分子mBD-2的表达,这可能与其防治肺部感染作用密切相关。Objective To study the effect of Buzhong Yiqi Decoction on the expression of mouse β-defensin 2（mBD-2） in lung epithelial cell.Methods C57BL/6 mice were randomly divided into control group and three experiment groups,in which mice were administered with different doses of Buzhong Yiqi Decoction.The mRNA and protein expression of mBD-2 were detected by RT-PCR and immunohistochemistry assay,respectively.Results The expression of mBD-2 could not be detected in control group.Treatment with Buzhong Yiqi Decoction significantly enhanced mRNA and protein expression of lung mBD-2 in mice,especially when higher dose of Buzhong Yiqi Decoction administrated.Conclusion Buzhong Yiqi Decoction might effectively upregulated mBD-2 expression in lung epithelial cells.The induction of effector molecule mBD-2 may be associated with the anti-infection effect of Buzhong Yiqi Decoction.
