摘 要:目的研究体外培养的人牙囊细胞的生物学特性。方法分离18岁人埋伏阻生下颌第三磨牙的牙囊,组织块法进行人牙囊细胞的培养,取不同代数细胞行HE染色观察细胞形态,MTS四唑盐比色法检测细胞增殖活性,茜素红染色定量分析成骨能力。结果第9代细胞面积比第3代、第6代细胞面积显著增大(P<0.05);第3、6、9代细胞增殖活性两两比较,差异有统计学意义(P<0.05);第3代细胞成骨能力明显强于第6代细胞(P<0.05)。结论随着体外培养代数的增加,人牙囊细胞生物学特征发生明显变化,面积逐渐增大,增殖活性逐渐变缓,成骨分化潜力逐渐减弱。Objective To study the characterication of human dental follicle cells (hDFCs)in vitro.Methods The dental follicles were dissecled from the unerupted lower third impacted molars,and then cultured in vitro.hDFCs of different passages were observed with microscopy,and their proliferation activities as well as osteoblast differentiation potential were evaluated.Results The hDFCs of 9th passage were significantly bigger than those of passage 3 and 6.We found among different passages,hDFCs of passage 3 appeared obviously greater osteoblasts differentiation potential than passage 6.Conclusion The characterization of hDFCs altered gradually cultured in vitro.
