摘 要:用 Eco R ,H ind ,Pst ,Bgl ,Bam H ,Xho ,Xba ,Sal 和 Kpn 共 9种限制性内切酶酶切对鲤鱼肝组织的 mt DNA进行酶解 ,利用 Gel- Pro Analyzer算出各酶切片段长度及mt DNA大小 ,测出其大小约为 1 6.61 kb( 1 kb=1× 1 0 3b) .另外 ,对电泳条件的优化进行了探讨 ,并将实验结果与以前报道的有关酶切实验结果进行了比较 ,表明鲤鱼 mt DNA的长度及限制性酶切位点均存在地域差异 .The mitochondrial DNA from liver of carp were digested by nine kinds of restriction endonuclease EcoRⅠ,HindⅢ,PstⅠ,BglⅡ,BamHⅠ,XhoⅠ,XbaⅠ,SalⅠ and KpnThe length of restriction fragments and the size of the mitochondrial DNA were calculated by using Gel Pro Analyzer. In the present study, the size of mtDNA of common carp is about 16.61 kb.The improvement of the electrophoresis conditions were also studied. The results of restriction fragments resulted from the present study were compared with those which were reported previously, which showed that there existed regional distinction in the size and restriction sites of mtDNA of common carp.
