胃痛消痞方对功能性消化不良大鼠血清及胃肠组织中CCK、VIP的影响

摘　　要：目的探讨胃痛消痞方对功能性消化不良(Functional dyspepsia,FD)肝郁脾虚型大鼠血清及胃肠组织中胆囊收缩素(Cholecystokinin,CCK)、血管活性肠肽(Vasoactive intestinal peptide,VIP)的影响。方法将40只大鼠随机分为正常对照组、模型组、西沙必利组及胃痛消痞方组,夹尾法将实验组造成FD大鼠模型后,每组分别给予相应的处理方案,利用ELISA检测血清CCK、VIP的含量,免疫组化法检测胃肠组织中CCK、VIP阳性纤维表达情况。结果模型组大鼠血清CCK含量及胃肠组织表达均减少(P<0.01),胃痛消痞方组CCK高于模型组(P<0.01)。模型组大鼠血清VIP含量及胃肠组织表达增加(P<0.01),胃痛消痞方组VIP低于模型组(P<0.01)。结论 FD发病可能与CCK减少、VIP增加等胃肠激素紊乱有关。胃痛消痞方通过调节血清CCK、VIP含量及两者在胃肠组织中的表达,改善胃肠运动功能,从而治疗肝郁脾虚型FD,且疗效优于西沙必利。Objective To observe the influence of weitongxiaopi decoction on serum and gastrointestinal cholecystokinin（CCK） and vasoactive intestinal peptide（VIP） in rats with functional dyspepsia（FD） of liver-depression and spleen-deficiency type.Methods All the rats were divided into 4 groups randomly：normal control group,model group,cisapride group,weitongxiaopi decoction group.After establishment of FD by tail clamp method,every group was given relevant measure.Serum and gastrointestinal CCK and VIP were examined by enzyme-linked immunosorbent assay（ELISA） and immuno-histochemical method respectively.Results Both the content of serum CCK and the expression of gastrointestinal CCK reduced in model group（P0.01）,and CCK of weitongxiaopi decoction group was higher than that of model group（P0.01）.Both the content of serum VIP and the expression of gastrointe-stinal VIP increased in model group（P0.01）,and the VIP of weitongxiaopi decoction group was lower than that of model group（P0.01）.Conclusion FD might be related to CCK decrease,VIP increase and other disordered gastrointestinal hormone.Weitongxiaopi decoction group can regulate CCK and VIP to some extent to cure the FD with incoordination between the liver and stomach,and it is superior to cisapride.
