摘 要:以弱酸类药物丙磺舒(Probenecid,PBD)在PAMPA模型和MDCK细胞模型上的累积吸收量、表观渗透系数(Apparent permeability coefficient,Papp)以及PBD跨细胞单层吸收对能量、pH的依赖性为指标,分析远曲小管上皮细胞对PBD的重吸收作用及其机制.结果发现在MDCK模型上PBD吸收是PAMPA上的3倍;并且PBD跨细胞吸收具有能量依赖性.提示肾远曲小管上皮细胞能主动重吸收弱酸类药物.In order to explore the reabsorption function of renal distal convoluted tubule cell, PAMPA model and MDCK monolayer were established and accumulative absorption of probenecid (PBD) was measured by these two models. The effects of culture temperature and pH on absorption of PBD cross MDCK monolayer were also studied in this paper. It was found that PBD absorption from MDCK model was 3 times more than that from PAMPA while DZP absorption was equal in these two models. And PBD absorption was temperature-dependent and pH- dependent at some degree. These indicated that both active transport and passive diffusion are involved in renal distal convoluted tubule reabsorption, but active transport is more important for weak acid drug at physiological pH.
