摘 要:从大鼠海马组织提取总RNA后,测定了不同时间和温度下保存及3次循环冻融后RNA的浓度和纯度,并分析了其完整性;探讨了保存时间、保存温度及反复冻融对大鼠海马组织总RNA质量的影响.结果表明,不同条件下保存及冻融3次后RNA浓度较首次检测值均有所降低,4℃下保存60d标本中的RNA浓度明显低于对照组(P<0.05),其他组未见显著性差异;各组RNA的OD 260/OD280在1.8~2.0之间.在-20℃和-70℃下分别保存30d和60d,以及24h冻融3次后的样品RNA的完整性较好;但4℃下保存的两组样品的RNA发生分解.RNA在4℃和-20℃下分别保存30d和60d后,相应的RE[(不同条件下保存后的样品浓度-初始浓度)/初始浓度]值均大于15%,而-70℃下保存30d、60d以及在24h内冻融3次后的RE值均小于15%.据此可知,大鼠海马组织总RNA样品可在-70℃下保存60d,反复冻融3次后总RNA质量基本不变.The total RNA was extracted from rat hippocampus. The concentration, purity and integrity of total RNA extracted from rat hippocampus were determined in relation to different preservation conditions including store time, store temperature and repeated freezing-thawing cycles. The effects of store time, store temperature and repeated freezing-thawing on the mass of total RNA in rat hippocampus were explored. It has been found that, as compared with the initially detected value, the concentration of RNA detected after storage in different preservation conditions and three freezing-thawing cycles all decreases. Significantly decreased RNA concentration is detected after storage at 4 ℃ for 60 d as compared with the control group (P 0. 05). No significant changes of RNA concentrations are observed in other preservation conditions, and the OD2G0/OD280 of RNA in various groups of tested samples is in the range of 1.8~2.0. Besides, no decomposition of RNA is observed in samples stored at - 20 ℃ and -70 ℃ for 30 d and 60 d or subjected to 3 freezing-thawing cycles (within 24 h), while the RNA in the two groups of samples stored at 4 ℃ is decomposed. Moreover, after the samples are stored for 30 d and 60 d at 4 ℃ and -20 , respectively, relevant RE values (the ratio of the difference between the concentration of the sample after storage and the initial concentra- tion to the initial concentration) are larger than 15 %, while those of the samples stored at -70 ℃ for 30 d and 60 d or subjected to 3 freezing-thawing cycles (within 24 h) are all lower than 15 %. This means that the total RNA of rat hippocampus samples can be stored at -70 ℃ for 60 d, and the total mass of RNA after 3 freezing-thawing cylces remains unchanged.
