膝骨痹康胶囊对膝骨性关节炎大鼠IL-6、IL-9影响的实验研究

摘　　要：目的探讨膝骨痹康胶囊对大鼠膝骨性关节炎(KOA)模型关节软骨中白细胞介素6(IL-6)和白细胞介素9(IL-9)水平的影响。方法选取60只清洁级SD大鼠,从中随机取10只作为空白对照组,其余采用单侧跟腱切除法制造大鼠KOA模型,将符合模型标准的大鼠随机分为五组:模型组,抗骨质增生胶囊组,膝骨痹康胶囊低、中、高剂量组。经药物干预30 d后,采用免疫组织化学方法观察膝关节软骨中IL-6和IL-9的表达。结果关节软骨中IL-6和IL-9的表达空白对照组与模型组比较差异有统计学意义(P<0.01);膝骨痹康胶囊中剂量组能显著下调关节软骨中IL-6和IL-9的表达,与模型组比较差异有统计学意义(P<0.01)。结论膝骨痹康胶囊能够减轻KOA关节软骨破坏及关节病损,其可能的机制是通过减少炎症细胞因子IL-6和IL-9的破坏作用来实现。Objective To explore the impacts of xigubikang capsules on the levels of interleukin - 6 （ IL - 6 ） and interleukin - 9 （ IL - 9 ） in the joint cartilage of the rat models with knee osteoarthritis （ KOA ）. Methods 60 clean - grade SD rats were selected. 10 rats of them were randomized to be in a blank control group. The rest rats were prepared as KOA models with unilateral Achilles tendon resection. The rats in compliance with the model standard were randomized into 5 groups, a model group, a kangguzhizengsheng capsules group, a low - dose xigubikang capsules group, a middle - dose xigubikang capsules group and a high - dose xigubikang capsules group. In 30 - day medicine intervention, the immunohistochemistry （IHC） method was adopted to observe the expressions of IL - 6 and IL - 9 in the knee joint cartilage. Results The difference in the expressions of IL -6 and IL -9 in the joint cartilage was significant statistically between the blank control group and the model group（ P 〈 0.01 ）. The expressions of IL- 6 and IL- 9 in the joint cartilage were down - regulated significantly in the middle - dose xigubikang capsules group, presenting the statistical significant difference as compared with that in the model group （P 〈 0.01 ）. Conclusion Xigubikang capsules alleviates the joint cartilage destruction and the joint damage in KOA, which is achieved probably through reducin the destruction of inflammatory cellular factors IL - 6 and IL - 9.
