摘  要:目的:观察人参皂甙对高能电子线一次大剂量照射荷瘤小鼠的影响。方法:6~8周健康雄性NIH小鼠,左后肢大腿外侧皮下接种小鼠S-180纤维肉瘤细胞或小鼠肝癌细胞,一周后选择肿瘤大小一致的小鼠40只,按是否接受高能电子线照射、给予人参皂甙方式等分为5组(8只小鼠/组)进行研究。接种后10d肿瘤局部高能电子线照射剂量为20Gy,人参皂甙腹腔注射剂量为50mg/kg。观察指标包括小鼠体重、肿瘤组织微循环变化、抑瘤率、生存率及肿瘤组织光电镜改变。结果:空白对照组:荷瘤存活期19~25d,瘤重(2.7±0.21)g,微血管直径15~20um,体重(30±2.83)g。单纯人参皂甙组:荷瘤存活期21~27d,瘤重(1.9243±0.2)g,抑瘤率28.39%(P<0.05),体重(31.0±1.85)g。单纯放疗组:荷瘤存活期25~34d,瘤重(0.68±0.13)g,抑瘤率74.88%,体重(27.9±1.89)g,微血管直径18~22um。照射前后均给予人参皂甙组:荷瘤存活27~37d,瘤重(0.86±0.1)g,抑瘤率67.91%,体重(32.1±2.3)g,血管直径15~20um。照射后给予人参皂甙组:瘤重(0.55±0.1)g,抑瘤率79.53%,体重(29.8±1.8)g,血管直径16~22um。实验结果提示人参皂甙用于实验小鼠肿瘤的放射治疗时,可以明显延长小鼠生存率,增加体重,对肿瘤放射治疗效果无明显影响。术前短时间给予人参皂甙可减少高能射线早期辐射损伤,其微循环改变较轻,恢复快。Objectives: To explore the effect of ginsenosdes on tumor bearing mice local irradiated by single high dose of high energy electrons. Methods: NIH male mice with left hind leg innoculated subcutaneously with murine S-180 cells or murine hepatic cancer celts, after seven days, selecting 40 cases of mice that the tumor size was consistent, they were divided into 5 groups (8 mice/group) according to the way of whether or not to accept the high energy electrons irradiation, and the ginsenosides, after 10 days vaccination, local tumor irradiation dose of high energy electrons for 20Gy and ginsenosides injected intraperitoneally dose of 50mg/kg. Outcome measures included body weight of mice, changes in the microcirculation of the tumor tissue, the inhibition rate, survival rate and tumor tissue photoelectric mirror changes. Results: The control group has a survival of 19-25 days, tumor weight was (2.7±0.21) g, Microvascular diameter was 15-20um, and bodyweight was (30-t-2.83) g. The pure ginsenosides group has a survival of 21-27 days, the weight of tumor was (1.9243 ±0.2) g, and the tumor inhibition rate was 28.39% (P〈0.05), body weight was (31.0±1.85) g. The pure radiation group has a survival of 25-34 days, the weight of tumor was (0.63±0.13) g, the tumor inhibition rate was 74.88%, and body weight was (27.9± 1.89) g. the microvascular diameter was 18-22um. Ginsenosides+radiation group has a survival of 27-37 days, the weight of tumor was (0.86-t-0.1) g, tumor inhibition rate was 67.91%, and body weight was (32.1±2.3) g, microvascular diameter was 15-20um, the tumor weight of after irradiation to give ginsenoside group was (0.55±0.1) g, the tumor inhibition rate was 79.53%, and body weight was (29.8±1.8) g, microvascular diameter was 16±22um. The preliminary results suggested that ginsenosides can significantly improve the survival of tumor, increase body weight, had no significant effect on the radiation oncology treatment.
