胰腺星状细胞在2型糖尿病胰岛纤维化进程中的影响

摘　　要：目的观察胰腺星状细胞（pancreatic stellate cell，PSC）在2型糖尿病（Dpe 2 Diabetes Mellitus，T2DM）大鼠胰岛中定植，探讨其在T2DM胰岛纤维化中的作用机制。材料方法分离、培养4月龄雄性GK大鼠（Goto-Kakizaki-GK）胰岛，24小时后继续培养胰岛，免疫荧光检测胰岛外生蛋白表达，如细胞波形蛋白（vimentin），α-平滑肌肌动蛋白（α-SMA）和胶原1（Col1）的表达；GK大鼠胰腺5μm连续冰冻切片，行Masson染色和免疫荧光检测（α-SMA，胰岛素（insulin），α-SMA／insulin）．对照组用性别年龄匹配Wistar大鼠。结果1．培养7d后，少许不规则鱼眼状细胞自GK大鼠胰岛内生长出，并特异性表达vimentin和α-SMA和Col 1：2．胰岛外生细胞在体外至少可以传代40代：3．GK大鼠胰岛形态大小不一，胰岛内Masson蓝染色胶原纤维明显增多；纤维化区域胰岛素分泌缺失，α-SMA染色阳性细胞增多。结论活化PSC定植于T2DM模型4月龄GK大鼠纤维化胰岛中，并可能在T2DM胰岛纤维化和胰岛B细胞功能衰竭中发挥了一定作用。Aims： The aim of the present study was to examine the role of PSC in the developmentof islets fibrosis in the spontaneous Type 2 Diabetes Mellitus （T2DM） model Goto-Kakizaki （GK） rat. Methods： Islets were iso- lated and cultured from 4-month-old-male-diabetic GK rat and age-matched Wistar control rat. Cells grown out from cultured islets were examined for vimentin, α -SMA and Col I by immunofluorescence microscopy. Masson staining assay, andimmunofluorescence microscopy 30 α-SMA, insulin, and α-SMA/insulin） were performed using pancreatic tissue from 4-month-old GK rat and age-matched Wistar rat. Results： 1. Cells grown out from islets of GK rat stained positive for α-SMA, vimentin and Coll. 2. these cells grown out from GK rat islet could be cultured at least 40 passages, in vitro; 3. Large disorganized and fibrotic islets are present in GK rat, insulin-/α -SMA＋ staining cells located in the fibrotic area of islets from GK rat as well as insulin staining positive cells decreased, as compared with that from Wistar rat. Conclusion： Our findings suggest that PSC presented in the islets of GK rat, and activated PSC may play an important role in the development of pancreatic islets fibrosis.
