摘 要:[目的]筛选肠道干细胞特征的差异蛋白质或蛋白质群。[方法]采用双向凝胶电泳方法分离新生小鼠肠边缘群细胞与肠上皮细胞总蛋白,选择差异表达超过1.5倍的蛋白质进行质谱分析及数据库搜索、鉴定。[结果]成功建立小鼠肠边缘群细胞与肠上皮细胞的双向电泳图谱,筛选出差异蛋白点39个,质谱分析及数据库搜索共鉴定出32种蛋白质。[结论]小鼠肠干细胞与上皮细胞的蛋白表达谱存在差异,鉴定出的32种蛋白质有可能成为潜在的、有应用价值的肠道干细胞鉴别分子标志。[Objective]To screen differential proteins or protein groups with stem cell characteristics.[Methods]Two-dimensional gel electrophoresis was used to separate total proteins from intestinal side population cells and intestinal epithelial cells of neonatal mice.Proteins which were differentially expressed more than 1.5 times were selected for mass spectrometry analysis as well as database retrieval and identification.[Results]The two-dimensional electrophoresis patterns of intestinal side population cells and intestinal epithelial cells were successfully established to screen 39 differential protein spots.Thirty-two kinds of proteins are identified using the mass spectrometry analysis and database retrieval.[Conclusion]There were differences between the protein expression profiles of mouse intestinal epithelial cells and stem cells.The identified 32 kinds of proteins may become potential and valuable molecular markers for identification of intestinal stem cells.
