摘  要:目的:探讨当归挥发油对增生性瘢痕成纤维细胞增殖、凋亡和胶原合成的影响。方法:体外培养人增生性瘢痕成纤维细胞,用噻唑蓝比色法检测细胞增殖活性,用流式细胞术分析细胞周期及凋亡,用放射免疫法测定胶原合成。结果:1mg/L、4mg/L和16mg/L当归挥发油组与对照组相比G0/G1期细胞、G2/M期减少,显著增加了S期细胞(P<0.05)。凋亡率随当归挥发油浓度增大逐渐增大(P<0.05)。当归挥发油呈剂量和时间依赖性抑制细胞合成胶原(P<0.05或0.01)。结论:当归挥发油抑制成纤维细胞的增殖和胶原合成。Objective To investigate the effects of Angelicanaphtha on proliferation cell cycle,apoptosis,and collagen synthesis of human hypertrophic scar fibroblast.Methods Human hypertrophic scar fibroblast was cultured and passaged in Dulbecco's modified Eagle's medium(DMEM) and treated with Angelicanaphtha 1mg/L,4mg/L,and 16mg/L at 1,3,5,and 7day respectively.The proliferation was measured with MTT method.The cell cycle and apoptosis were analyzed with Flow cytometry and collagen synthesis was determined with radioimmunoassay.Results Angelicanaphtha of 1mg/L,4mg/L,and 16mg/L made G0/G1 phase cells,G2/ M phase cells decrease and S phase cells remarkable increase(P0.05).Furthermore,apoptosis of the fibroblasts was increased with increase of Angelicanaphtha concentration(P0.05).The collagen synthesis of fibroblast was inhibited by Angelicanaphtha in concentration-dependent manner(P0.05).Conclusion The proliferation and collagen synthesis of fibroblast was inhibited by Angelicanaphtha..
