摘 要:目的 研究丹参酮Ⅱ A 磺酸钠对缺血/ 再灌注(I/R)模型大鼠脑梗死体积、脑含水量及神经功能 评分的影响.方法 40 只SD 大鼠按随机数字表法分成假手术组、模型组、丹参酮Ⅱ A 预处理组、丹参酮Ⅱ A 治 疗组,每组10 只。用改良Longa 栓线法复制大脑中动脉闭塞(MCAO)模型,2 h 后开放血流再灌注。24 h 处死 大鼠取脑。比较分析各组大鼠脑梗死体积(%)、脑含水量(%)及术后12 h 和24 h 神经功能评分. 结果 模型 组脑梗死体积明显高于丹参酮Ⅱ A 预处理组和治疗组〔(33.5±6.7)% 比(24.3±4.8)%、(25.3±4.6)%,均P < 0.01〕;模型组脑含水量明显高于预处理组和治疗组〔(84.6±4.2)% 比(79.9±6.3)%、(80.1±6.0)%,均P <0.01〕; 模型组12 h、24 h 神经功能评分(分)高于预处理组和治疗组(12 h :3.10±0.48 比2.30±0.59、2.50±0.50,24 h : 3.30±0.62 比1.90±0.36、2.10±0.46,P <0.05 或P <0.01).结论 丹参酮Ⅱ A 磺酸钠可以减少I/R 模型大鼠 的脑梗死体积,降低脑含水量,改善神经功能评分.Objective To study the effects of tanshinone Ⅱ A sulfonate on cerebral infarct volume, brain water content and neurologieal functional score in rat models with focal cerebral ischemia/reperfusion (I/R). Methods Forty Sprague-Dawley (SD) rats were randomly divided into four groups based on the random number table : sham operation group, model group, tanshinone Ⅱ A sulfonate pretreatment group and tanshinone Ⅱ A sulfonate treatment group (each, n =10). The rat model of I/R was reproduced by application of modified Longa suture method to induce middle cerebral artery occlusion (MCAO), and after 2 hours, the reperfusion was pertormed by opening the blood flow. The rats were executed and their brains removed after 24 hours. The cerebral infarction volume (%), brain water content (%), and neurological functional score of the rats at 12 hours and 24 hours after reperfusion were analyzed in each group and compared among them. Results In the model group, the percentages of infarct volume and the brain water content were significantly higher than those in the tanshinone ⅡA sulfonate pretreatmem and treatment groups ( infarct volume : ( 33.5 ± 6.7 ) % vs. ( 24.3 ± 4.8 ) %, ( 25.3 ± 4.6 ) % ; brain water content : (84.6±4.2)% vs. (79.9±6.3)%,(80.1±6.0)%, all P〈0.01] ; the neurological functional scores of model group at different time points were obviously greater than those in the pretreatment and treatment groups ( 12 hours: 3.10±0.48 vs. 2.30±0.59, 2.50±0.50; 24 hours: 3.30±0.62 vs. 1.90±0.36, 2.10±0.46, P〈0.05 orP〈 0.01). Conclusion Tanshinone Ⅱ A sulfonate can reduce cerebral infarct volume, brain water content and improve neurological functional score of rat models with I/R.
