摘 要:目的建立SD大鼠和Wistar大鼠同种异体卵巢皮下游离移植模型,并以Wistar大鼠自体卵巢皮下游离移植模型作为对照组,以补肾安胎中药复方"寿胎丸"进行干预,观察移植模型凋亡相关基因Fas、FasL的表达,探讨"寿胎丸"抗卵巢移植急性排斥反应的机理。方法 210只Wistar雌性大鼠随机分为7组,即自体移植模型组、自体移植+寿胎丸组(均P<0.05)、异体移植模型组、异体移植+寿胎丸组、异体移植+CsA组、异体移植+CsA+寿胎丸组和正常对照组。各组大鼠于移植术后7、14、21、28、35d分批处死,每次6只。取左侧卵巢,石蜡包埋切片,免疫组化检测凋亡分子Fas及其天然配体FasL的表达。结果移植术后14 d,自体移植模型组的Fas水平低于正常对照组(P<0.05),其余各时间点,自体移植模型组、异体移植模型组与正常对照组大鼠比较,均无明显差异(P>0.05);移植术后28d,异体移植+CsA组的Fas水平低于异体移植模型组和异体移植+寿胎丸组(均P<0.05),异体移植+CsA+寿胎丸组Fas水平低于异体移植+寿胎丸组(P<0.05)。比较各组移植卵巢内的FasL水平,在各时间点均无统计学差异(P>0.05)。结论寿胎丸对Fas、FasL的调控作用不明显,可能与卵巢内的Fas、FasL低表达有关。Objective Establish allograft ovary subcutaneous free transplantation model with SD rats and Wistar rats, and set autologous skin free transplantation model with Wistar rats as the control group. Using the recipe of "Shoutai Wan" which has the function of invigorating the kidney and preventing miscarriage to these rats, observe the expression of the apoptotosis related genes -Fas,FasL, to explore the possi- ble mechanisms of anti acute rejection of ovary transplantation of the Shoutai Wan. Methods 210 female Wistar rat were randomly divided into the following seven groups(30 in each group): Autologous transplantation group,autologous transplantation + Shoutai Wan group, al- lograft transplantation group,aUograft + Shoutai Wan group, allograft + Cielosporin A (CsA) group,aUograft + CsA and Shoutai Wan group, normal controls. On the d7,d14,d21 ,d28,d3] after the transplantation operation, sacrificed 6 rats in each group and collected the left ovary, detecting the expression of apoptosis related genes Fas/FasL. Results The examination of Fas expression of transplanted ovaries indi- cated that Fas level of Autologous transplantation group was much lower than normal controls (P 〈 0.05 ) 14 days after the transplantation, and there were no obvious differences among the three groups in other time(P 〉0.05). 28 days after the transplantation, the Fas level of allograft + CsA group was much lower than that of allograft transplantation group and allograft + Shoutai Wan group( P 〈 0.05 ), and Fas level of al- lograft + CsA and Shoutai Wan group was much lower than that of allograft + Shoutai Wan group (P 〈 0.05 ). The examination of FasL ex- pression indicated that The FasL level of seven groups didn't have significant difference at any time after the transplantation(P 〉 0.05 ). Con- clusion ShouTai Wan doesn't have manifested function of mediating the expression of Fas,FasL,may be relate to the? lower expression in the ovaries.
