摘  要:目的:观察Visfatin在人增殖性糖尿病视网膜病变(PDR)病人纤维血管膜中的表达,探讨其在PDR中可能的作用机制。方法:收集16例行玻璃体手术的增殖性糖尿病视网膜病变病人术中取出的纤维血管膜(Fibrovascular membranes,FVMs),应用免疫荧光方法检测Visfatin和GFAP的蛋白表达及定位,TR-PCR检测Visfatin在纤维血管膜中mRNA表达。结果:Visfatin免疫荧光结果显示在所有16例血管膜中均有强阳性表达且与GFAP共表达,RT-PCR显示在检测的8例中7例强表达。结论:Visfatin在PDR病人FVMs的表达可能与增殖性糖尿病视网膜病变纤维血管膜的形成相关,Visfatin可能成为治疗PDR的新靶点。Objective:To investigate the expression of visfatin in human fibrovascular membranes (FVMs) of proliferative diabetic retinopathy (PDR) patients, and try to explore the possible role of visfa- tin in the occurrence and development of PDR. Methods: FVMs samples were collected from 16 PDR patients by vitrectomy. Visfatin and GFAP expression in the membranes were examined by immunofluo- rescence analyses;total mRNA was isolated from the samples and detected by RT -PCR. Results: Im- munofluorescence analysis showed that visfatin and GFAP were co - expressed in most of the ceils in the FVMs and visfatin mRNA was detectable. Conclusion: Visfatin was detectable at both protein and mRNA levels in all excised fibrovascular membranes, which suggests that it may play a role in the path- ogenesis of fibrovascular membranes in PDR patients.
