摘  要:目的探讨吡那地尔预处理对缺氧/复氧后心肌细胞钙敏感性受体(calcium-sensingreceptor,CaSR)的影响。方法通过离体大鼠心脏灌注装置,分离成年大鼠心肌细胞随机分为四组,其中正常组(N组)常规培养,另三组缺氧45min、复氧60min制作缺氧/复氧模型;C组不做预处理;吡那地尔组(P组)和格列苯脲组(G组)在缺氧前用吡那地尔30μmol/L预处理15min;G组在吡那地尔处理前用格列苯脲50μmol/L处理5min。采用RT-PCR及Westernblot方法检测心肌细胞CaSR基因及蛋白表达情况。结果 C、P、G组CaSR基因表达水平及蛋白表达量明显高于N组(P<0.01);P组CaSR基因表达水平及蛋白表达量明显低于C组(P<0.05);而G组明显高于P组(P<0.01)。结论吡那地尔预处理能抑制心肌细胞缺氧/复氧导致的CaSR基因表达上调,其作用可能与ATP敏感性钾通道的开放有关。Objective To investigate the effect of pinacidil preconditioning on calcium-sensing receptor (CaSR) expression of ventricular myocytes after hypoxia/re-oxygenation, in rats. Methods Adult rat cardiomyocytes were isolated by heart perfusion apparatus and randomly divided into the following four groups: normal group (group N), control group (group C), pinacidit preconditioning group (group P) and glibenclamide group (group G). The cells in group N were normally cultured. In the other three groups, oxygen was deprived for 45 min followed by reoxygenation for 60 min. Groups P and G were pretreated with 30 μmol/L pinacidil for 15 min before hypoxia. Group G was treated with 50 μmol/L glibenclamide for 5 min before pinacidil preconditioning. The expressions of CaSR mRNA and protein were detected by the Real-Time PCR and Western blot, respectively. Results The expressions of CaSR mRNA and protein in group N decreased compared with other three groups (P〈0.01). When compared with group P, CaSR expressions in groups C and G were increased (P〈0.05). Conclusion Pinacidil preconditioning can inhibit hypoxia/reoxygenation induced CaSR expressions, which might be mediated by the opening of ATP sensitive potassium channel.
