摘 要:XJ 16 0病毒是我国首次分离的辛德毕斯病毒 ,在对其全基因组核苷酸序列测定的基础上 ,对病毒基因组序列、病毒同源性和病毒系统进化等进行了较为详细的分析 ,结果表明 :XJ 16 0病毒具有典型的甲病毒属辛德毕斯病毒的序列特征。该病毒nsp3蛋白基因中有 11处缺失及两处插入 (45 17~ 5 485 ) ,涉及 90个核苷酸。核苷酸序列的系统进化分析表明 ,XJ 16 0病毒属于辛德毕斯病毒欧洲 /非洲亚组。病毒全基因组核苷酸序列进化分析显示 ,XJ 16 0病毒是一株新的甲病毒或辛德毕斯病毒的新亚型。A phylogenetic analysis and a distance matrix tree based on the full length of genome of XJ 160 virus has been determined in this study. XJ 160 conserved all consensus sequence elements commonly found in alphaviruses. Compared with Sindbis virus AR339, significant divergence was found in the XJ 160 sequence. Globally, XJ 160 shares only 82% nucleotide and 91.4% amino acid identity with the AR339 strain. The divergence in nucleotides and amino acids between the two viruses is nearly three times of those of any other two sindbis viruses. Moreover, 11 deletions and insertions are found in the nsP3 region (4?517-5?485) involving 99 nucleotides. We performed a phylogenetic analysis based on 3′ nontranslated region sequence,it shows XJ-160 is closer to the African/European group than to Asian/Australian group. The another phylogenetic tree based on the full-length genomic sequences of seven Sindbis-like viruses currently available in GenBank database, XJ 160 forms a separate grouping in this tree. These results suggest that XJ 160 is a new member of the Sindbis family.
