摘  要:目的:研究大孔树脂分离纯化积雪草总苷过程,优化相关工艺条件和参数。方法:考察HPD100、HPD200、HPD500、AB-8、D101五种大孔树脂对积雪草总苷的动态、静态吸附量和解吸率选定型号,以积雪草总苷为指标考察纯化工艺参数。结果:优选确定AB-8型大孔树脂,上样生药质量浓度为0.0625 g/ml,上样量中生药与树脂比为1:2(g:ml),洗脱剂为70%乙醇,洗脱体积为2BV,验证试验平均洗脱率为81.27%,洗脱后积雪草总苷纯度为55.27%。Objective:To study the purification process of total glycosides from Centella asiatica by Macroporous Resins,optimizing the process conditions and parameters.Methods: To find the appropriate macroporous resin model by Inspecting the dynamic,static adsorption and desorption rate of total glycosides of Centella asiatica with five kinds macroporous resin(HPD100,HPD200,HPD500,AB-8,D101),investigated the purification process parameters by total glucosides of Centella asiatica as index.Results: Determined AB-8 macroporous resin by optimizing,the crude drug’s concentration is 0.0625g/ml,the ratio of crude drug and resin is 1:2(g:ml),the eluent is 70% ethanol and elution volume for 2BV.The recovery was 81.27% and the purity reached 55.27%.Conclusion: AB-8 macroporous resin is suitable for separation and purification of total glucosides from Centella asiatica.
