摘 要:采用胎盘绒毛组织体外孵育模型 ,观察了胎盘阿片肽对人早期胎盘滋养层组织基质金属蛋白酶 2和 9活力的调节。结果表明 ,β 内啡肽 (10 -9mol/L)和强啡肽 (10 -10mol/L)可以显著地抑制体外孵育的人早期胎盘滋养层组织中这两种金属蛋白酶的活力 ,且其抑制效应被纳洛酮 (10 -8mol/L)所阻断 ;甲硫氨酸脑啡肽在 10 -6mol/L时显著提高二种基质金属蛋白酶的活力 ,而在 10 -8~ 10 -10 mol/L时对基质金属蛋白酶的活力无明显效应 ;亮氨酸脑啡肽则在所有试验浓度条件下对基质金属蛋白酶的活力都无显著影响。提示胎盘阿片肽可通过其受体影响基质金属蛋白酶的活力水平 。Opioid peptides can be synthesised by placenta and act as endocrine and paracrine factors in placenta. Matrix metalloproteinases(MMPs) also can be synthesised and secreted by placenta and take an important role during implantation. In the present study, trophoblast tissues from human early placenta were incubated to study the opioid peptides regulation of MMPs. Analysis of medium by zymography determinated activities of MMP 2(EC 3. 4. 24. 24) and MMP 9(EC 3.4.24.35). β endorphin and dynorphin significantly decreased the activities of MMP 2 and MMP 9 in incubation medium ( P <0 01), and naloxone block these inhibition effects of β endorphin and dynorphin. Leu enkephalin had no effect on the activities of MMP 2 and MMP 9, Met enkephalin at 10 -6 mol/L significantly increased activities of MMP 2 and MMP 9 ( P <0 01) but had no significant effect at concentrations below 10 -6 mol/L. These data suggest that opioid peptides take a role in implantation through its regulation in MMP 2 and MMP 9 activities of human trophoblast.
