摘  要:目的:探讨白细胞介素-1B基因多态性与甲型H1N1流感温邪传变模式的关系。方法:建立基于飞行时间质谱技术的SNP分型方法,检测167例甲流患者和192例健康对照人群IL-1B基因rs1143623和rs16944位点多态性,将甲流患者辨证分型为温邪不传、顺传和逆传3种模式,比较不同人群等位基因频率差异。结果:rs16944位点具有A和G两种等位基因,A等位基因频率在健康对照、不传型、顺传型和逆传型人群分别为49.5%、60.2%、62.5%和71.4%,甲流3种传变模式人群A等位基因频率均显著高于健康对照(P=0.016,P=0.013,P=0.025)。rs1143623位点等位基因频率在不同人群中差异无统计学意义(P>0.05)。结论:IL-1B基因rs169441位点多态性与甲型H1N1流感温邪传变模式相关,A等位基因为易感基因,可以作为甲流中医辨证分型的分子遗传标签。Objective To explore the association between IL-1B gene polymorphisms and warm pathogen transmission model of A H1N1 influenza.Methods The genotypes of SNP(rs1143623,rs16944) in IL-1B gene were determined in H1N1 group(n=167) and healthy control group(n=192) with TOF-MS assay,and H1N1 group was divided into 3 transmission models involving No-transmission,For-transmission,and Rev-transmission,then allelic frequency was compared among different groups.Results rs16944 SNP involved A and G allele,A allele frequency was 49.5%,60.2%,62.5%,and 71.4% in controls,No-transmission,For-transmission,and Rev-transmission groups respectively,and it was higher in 3 transmission models than in controls(P=0.016,P=0.013,P=0.025).For rs1143623 SNP,no significant difference was observed in allele frequency among different groups.Conclusion IL-1B gene rs16944 SNP was associated with warm pathogen transmission model of A H1N1 influenza,as susceptible allele,T allele could be considered as a new molecular marker for syndrome differentiation in TCM of A H1N1 influenza.
