摘 要:目的探讨三七总皂苷(PNS)治疗百草枯(PQ)中毒肺损伤的效果及机制。方法将150只SD雄性大鼠分为对照组(30只)、PQ组(60只)及PNS组(60只)。PQ组和PNS组一次性予PQ 25 mg/kg灌胃染毒,制作PQ中毒模型,对照组予等体积生理盐水灌胃。PNS组于染毒前15 min阴茎静脉注射PNS 50 mg/kg,以后每日1次给药直至处死前;PQ组、对照组分别在同时间点阴茎静脉注射等体积生理盐水。观察各组染毒后6 h、12 h、1 d、3 d、5d、7 d肺组织病理改变,检测血清IL-1β水平、肺泡灌洗液中PMN的NF-κB p65表达和中性粒细胞凋亡率。结果对照组肺组织结构基本完整,PQ组炎性细胞浸润明显增多,肺组织损伤程度加重,PNS组肺组织损伤程度较轻。PQ组IL-1β水平与NF-κB p65水平明显高于对照组、明显低于PNS组(P均<0.01)。2 d时PQ组PMN凋亡率明显低于对照组,PNS组相应时间点明显高于PQ组(P均<0.05)。结论三七总皂苷能减轻百草枯中毒导致的肺组织损伤,其机制为降低肺组织中IL-1β与NF-κB水平。Objective To investigate the effect of panax notoginseng saponins(PNS) on the paraquat poisoned lung in- jury and its mechanism. Methods Total of 150 male SD rats were randomly divided into 3 groups: normal control (C) group (30 rats ), PQ group (70 rats) and PNS group (70 rats). The PQ and PNS groups were given paraquat by gastric per- fusion(25 mg/kg) ; The C group was given saline by gastric perfusion. The PNS group was protected by intravenous injec- tions of penis with PNS(50 mg/kg) 15 mins before exposure of paraquat, once a day until killed. The PQ and C groups were given saline. Histological changes of lung tissues were observed at different time of 6 h, 12 h, 1 d, 3 d and 7 d after poisoned. The level of IL-1β in serum, the expression of NF-κB P65 in PMN of BALF and the ratio of PMN apoptosis were detected. Results The lung tissue structure of C group was basically integrate, and which of the PQ group showed more infiltration of inflammatory cells and serious injury, the injury of the PNS group was slighter. The level of IL-1β and NF-κB p65 in PQ group were significantly higher than those in C roup and lower than those in PNS group( all P 〈 0.01 ). The ratio of PMN apoptosis at 2 d in PQ group was lower than those in C group and PNS group( all P 〈 0.05 ). Conclusions PNS may alleviate lung injury in acute paraquat poisoned rats by inhibiting the expression of IL-1β and NF-κB P65 .
