摘  要:目的:建立测定雷公藤药材中雷公藤甲素含量的方法,比较分析不同产地雷公藤药材的质量。方法:采用高效液相色谱法,使用C18色谱柱(4.6 mm×250 mm,5μm),流动相A为甲醇,流动相B为水,采用梯度洗脱,于218nm处检测,测定雷公藤甲素的含量。结果:高效液相色谱法测定雷公藤甲素浓度在39~195μg.mL-1范围内线性关系良好,r=0.999 9,平均加样回收率为96.45%,RSD为3.19%(n=6)。结论:该方法准确可靠、简便、重复性好,可用于雷公藤药材质量分析控制,不同产地的雷公藤药材中雷公藤甲素的含量差异较大。Objective: To set up the process of assaying triptolide in tripterygium wilfordii in different habitat with high performance liquid chromatography (HPLC), and compare the quality of tripterygium wilfordii in different habitat. Methods: Triptolide was determined in 218 nm wave -length with high performance liquid chromatography (HPLC) and C18 chromatographic column (4. 6 mmx 250 mm, 5μm). Mobile phase A was methanol, and mobile phase B was water. Gradient elution was adopted. Results: In the range of 39 -195 μg·mL- 1 with HPLC, the linear relationship of triptolide was good, r = 0. 999 9, the average spotting recovery rate was 96. 45 %, and RSD = 3.19 % ( n = 6). Conclusion : This process is accurate, sensitive, reproducible, simple and easy to operate, and can be used as the quality control method for triptolide in tripterygium wilfordii in different habitat. Triptolide. The content of triptolide in tripterygium wilfordii in different habitat is different markedly.
