乙酰乳香酸抑制日本血吸虫虫卵肉芽肿形成及其机制的探讨

摘  要:目的探讨乙酰乳香酸(acetyl boswellia acid,ABA)抑制日本血吸虫虫卵肉芽肿形成及其作用机制。方法 6w龄SPF级C57鼠随机分成5组:A组为未感染血吸虫尾蚴同期正常鼠,B、C、D和E 4组经腹部皮肤感染日本血吸虫尾蚴20±2条。小鼠感染尾蚴21d后,B组经腹腔注射溶剂环糊精280mg/(kg.d)×20d,C组感染后不加治疗,D和E组分别腹腔注射ABA 140mg/(kg.d)×20d、280mg/(kg.d)×20d。所有小鼠12w龄时处死取血收集血清,转氨酶试剂盒检测血清ALT、AST;ELISA法检测血清白三烯B4(LTB4)。取小鼠肝组织中叶组织固定、切片做苏木素-伊红(HE)染色,观察并测量肝虫卵肉芽肿面积,计算与正常肝组织的比值。结果 E组ALT,AST和LTB4的水平均显著低于B和C组(P<0.05),D组ALT,AST和LTB4的水平虽均低于B和C组,但无统计学差异(P>0.05)。E组虫卵肉芽肿面积与正常肝组织的比值均显著小于B和C组(P<0.05),D组虫卵肉芽肿面积比值虽均小于B和C组,但无统计学差异(P>0.05)。结论 ABA对抑制血吸虫虫卵肉芽肿炎症有一定效果,实验结果说明ABA可降低LTB4的生成,是抑制肉芽肿炎症反应的机制之一。The purpose of this study is to investigate effect of acetyl boswellia acid(ABA) on formation of granulation induced by Schistosoma japonicum egg and its mechanism of action.Six-weeks old SPF level C57 mice were divided randomly into 5 groups: Group A was served as control;Groups B,C,D and E were infected with Schistosoma japonicum cercariae(20 ± 2).In 21th day post-infection,mice in Group B was given injection of solvents cyclodextrin intraperitoneally(280 mg/(kg · d) × 20 d);Group C without any treatment;Groups D and E were respectively given intraperitoneally ABA 140 mg/(kg · d) × 20 d,280 mg/(kg · d) × 20 d.All the mice were executed after 12 w and the serum was collected.Serum ALT and AST were detected by transaminase kit and serum leukotriene B4(LTB4)was detected by ELISA kit.Middle lobes of liver tissue of mice were fixed and prepared to sections for hematoxylin-eosin(HE) staining,and the sizes of egg granuloma of the liver were observed.Results showed that levels in ALT,AST and LTB4 in Group E were significantly lower than those in Groups B and C(P0.05),and the levels of ALT,AST and LTB4 in Group D were also lower than those in Groups B and C,but no statistically significant difference(P0.05).The size of egg granuloma in Group E was significantly smaller than that in Groups B and C(P0.05).The size of egg granuloma in Group D was smaller than that in Groups B and C,but no statistically significant difference(P0.05).In conclusion,one of the mechanisms of inhibition on granulomatous inflammation,the suppression from ABA to the generation of LTB4,has certain effects of inflammation in Schistosoma japonicum egg granuloma.
