摘  要:目的调查新疆博乐地区恙虫病东方体自然疫源地情况。方法采用捕鼠取脾,鼠体表捕螨,当地牧民静脉取血等收集样本,用试剂盒提取样本DNA,应用巢式PCR(nPCR)检测东方体基因(Sta56)。结果在博乐地区平原湿地区捕鼠126只,其中7只nPCR检测为阳性(阳性率5.55%);从当地牧民300份血液中nPCR检测6份为阳性(阳性率2.0%);野鼠体表带螨率为8.4%,从捕获博乐纤恙螨中nPCR检出阳性。DNA序列分析表明,鼠类、人血及恙螨中检出的基因序列相同,与东方体Karp株Sta56基因序列同源性为99%。在山地草原捕获鼠58只和捕获旱獭26只,nPCR检测其样本均为阴性,从鼠体表未捕获到恙螨。结论新疆博乐地区平原湿地存在恙虫病东方体自然疫源地,小家鼠、普通仓鼠、大沙土鼠为其宿主动物,纤恙螨为当地恙虫病的传播媒介。The aim was to investigate natural epidemic focus of Orientia tsutsugamushi(O.tsutsugamushi) in Bole area of Xinjiang,China.DNA was extracted from spleens of the captured mice and the blood of the local herdsmen,and the chigger mite were seized from the rodent body surface.Nested polymerase chain reaction(nPCR) technique was used to detect the Ot-Sta 56 gene.The positive samples were sequenced and analyzed by NCBI and analyzed in BLAST.Positive results were detected from all of the rodents,local herdsmen and chigger mite.O.tsutsugamushi were isolated from the rodent's spleen and trombidiid.The prevalence of Ot-Sta 56 gene was 5.55% and 8.40% for the blood and body surface of mice,respectively.The Sta56 gene nucleotide sequence homology to Karp strain was 99%.It's suggested that the natural epidemic focus of O.tsutsugamushi could exist in plain and wetland in Bole area.The host could be M.musculus,C.migratorius,and R.opimus,and the medium could be L.bolei.
