摘  要:目的:观察糖皮质激素肾阴虚证模型各靶组织相关激素受体的表达变化以及滋肾阴方的干预作用。方法:实验分为4组,正常组、模型组、模型+滋肾阴复方组,正常+滋肾阴复方组。模型组、模型+滋肾阴复方组采用氢化可的松腹腔注射50mg/kg,连续7天,造成大鼠肾阴虚证模型。采用Western blot检测各组大鼠下丘脑、胸腺、性腺的糖皮质激素受体α(GRα)、甲状腺激素受体α/β(TRα/β)、雄激素受体(AR)的表达。结果:糖皮质激素造模导致大鼠体重降低、肾上腺、胸腺重量降低;也致GRα、TRα/β、AR的表达改变,使GRα表达在下丘脑下调,在胸腺上调。滋肾阴方使用后,能显著保护动物体重、胸腺重量,肾上腺重量有明显上升趋势;并能显著降低GR在胸腺的表达,升高AR在性腺的表达,升高TRα/β在胸腺的表达。结论:糖皮质激素肾阴虚模型相应靶组织受体表达也发生了显著改变,在研究补肾中药时,需要注意研究其在受体和受体后水平的效应,而不仅仅关注对血浆激素水平的影响。Objective:To investigate the expressions of several hormone receptors in tissues in kidney -yin deficicency model caused by glucocorticoids. The effect of kidney - yin - tonifying formula was also studied. Methods : Rats were as- signed into 4 groups : normal, model, model plus kidney - yin -tonifying formula, and normal plus kidney - yin - tonifying formula group. Rats were administrated with peritoneal injection of cortisol at dose of 50mg/( kg . day)to generate kidney - yin deficiency model. At the end of experiment, the body weight, the weight of thymus, adrenal gland, liver and kidney were measured. The GRot,TRcx/~ and AR protein expressions in hypothalamus, thymus and gonad gland were detected with Western blot. Results:Glucocorticoids resulted in significant decrease in body weight, adrenal gland and thymus weight. Glucocorticoids down- regulated the GRot in hypothalamus ,up- regulated in thymus. After treatment by kidney - yin - tonifying formula, the body weight decreased, thymus weight was increased. The GRc~ expression in thymus was down- regulated,the AR in gonad was up -regulated, while the TRα/βin thymus was up -regulated. Conclusion:The protein expressions of hormone receptors in tissues in glucocorticoids - induced kidny - yin deficiency model were signifi- cantly changed. The effects of kidney - yin - tonifying formula at receptor and/or post - receptor level should be noted.
