摘  要:目的探讨聚氨酯、硅胶、聚氯乙烯3种中心静脉导管医用生物材料对XWLC-05(Xuanwei LungCancer-05)细胞增殖、凋亡及细胞周期的影响,为临床选择中心静脉导管提供依据。方法取XWLC-05细胞系复苏、培养并传代,取第3代细胞分别与大小为1.0 cm×1.0 cm的聚氨酯、硅胶、聚氯乙烯培养,以不加材料培养作为对照。培养72 h时倒置显微镜下观察细胞生长情况;培养24、48、72 h,MTT法检测细胞增殖,采用流式细胞仪检测细胞周期及凋亡率。结果倒置显微镜下观察示,培养72 h对照组细胞生长良好,聚氨酯组与硅胶组细胞生长情况与对照组相似;聚氯乙烯组细胞数量减少,坏死、溶解,残留贴壁细胞形态畸形,透光度降低。MTT法检测3种材料与细胞共培养24、48 h,细胞增殖、细胞周期及凋亡率与对照组比较,差异均无统计学意义(P>0.05)。72 h时与对照组比较,各实验组材料均显著抑制细胞增殖(P<0.05),其中聚氯乙烯组较硅胶组及聚氨酯组明显(P<0.05),硅胶组及聚氨酯组间差异无统计学意义(P>0.05);各实验组材料均显著影响细胞凋亡率和细胞周期,其中聚氯乙烯组显著,硅胶组次之,聚氨酯组最小,组间比较差异均有统计学意义(P<0.05)。结论聚氯乙烯显著影响XWLC-05细胞的增殖、凋亡及细胞周期;与聚氯乙烯、硅胶相比,聚氨酯材料具有较好生物相容性。Objective To explore the influence of three central venous catheter biomedical materials(polyurethane,silicone,and polyvinyl chloride) on the proliferation,apoptosis,and cell cycle of Xuanwei Lung Cancer-05(XWLC-05) cells soas to provide the basis for clinical choice of central venous catheter.Methods XWLC-05 cells were cultured and subcultured,and the cells at passage 3 were cultured with polyurethane,silicone,and polyvinyl chloride(1.0 cm × 1.0 cm in size),and onlycells served as a control.At 24,48,and 72 hours after cultured,MTT assay was used to detect the cellular proliferation and flowcytometry to detect the cell cycle and apoptosis.At 72 hours after cultured,inverted microscope was used to observe the cellgrowth.Results Inverted microscope showed the cells grew well in control group,polyurethane group,and silicone group.In polyvinyl chloride group,the cells decreased,necrosed,and dissolved;residual adherent cells had morphologic deformityand decreased transmittance.At 24 and 48 hours,no significant diff erence in proliferation,apoptosis,and cell cycle was foundamong 4 groups(P〈0.05).At 72 hours,the proliferations of XWLC-05 cells in three material groups were significantly inhibitedwhen compared with control group(P〈0.05),and the cells in polyvinyl chloride group had more significant proliferationinhibition than polyurethane group and silicone group(P〈0.05),but there was no signifcant difference in proliferationinhibition between polyurethane group and silicone group(P〉0.05).Compared with the control group,three material groupshad significant impact on the rate of apoptosis and cell cycle: polyvinyl chloride group was the most remarkable,followed bysilicone group,polyurethane group was minimum(P〈0.05).Conclusion Polyvinyl chloride can significantly impact theproliferation,apoptosis,and cell cycle of XWLC-05 cells;polyurethane has better biocompatibility than polyvinyl chloride andsilicone.
