摘  要:目的研究丹酚酸B对大鼠脑缺血再灌注(I/R)后缺血侧纹状体氨基酸类神经递质的影响,探讨丹酚酸B的脑保护作用机制。方法采用LONGA EZ法制备SD大鼠大脑中动脉栓塞(MCAO)模型。造模当天(第1天)腹腔注射丹酚酸B 1 mg/kg和丹酚酸B 10 mg/kg,每天1次,连续28 d。分别于给药第3天和第28天收集清醒大鼠缺血侧纹状体透析液,高效液相色谱-荧光检测法(HPLC-FL)测定谷氨酸(Glu)和γ-氨基丁酸(GABA)含量。结果大鼠脑缺血再灌注损伤后第3天,模型组Glu含量显著高于假手术组,GABA含量有升高趋势,丹酚酸B对Glu、GABA含量无显著影响。第28天,模型组Glu、GABA含量均保持在较高水平,丹酚酸B小剂量组、丹酚酸B大剂量组Glu、GABA含量均显著降低,与模型组比,P<0.05。结论随脑缺血时间延长,丹酚酸B可显著降低脑缺血再灌注损伤大鼠纹状体Glu、GABA含量,推测其脑保护作用可能与调节Glu、GABA含量有关。Objective To study the influences of salvianolic acid B(SalB) on ischemia striatal amino acid neurotransmitters in rats with cerebral ischemia/ reperfusion(I/R) injury,and discuss the mechanism of SalB in brain protection.Methods The model of middle cerebral artery occlusion(MCAO) was established in SD rats by using Longa EZ method.In the day of modeling(the first day) SalB(1 mg/kg and 10 mg/kg) were intraperitoneally injected to rats once a day for 28 days.On the 3rd day and 28th day the dialysate in ischemia striatum were collected from awake rats respectively.The content of glutamic acid(Glu) and γ-aminobutyric acid(GABA) were detected by using HPLC-fluorescent assay(HPLC-FL).Results On the 3rd day,Glu content was significantly higher in model group than that in sham-operation group,and GABA content had a increasing trend.SalB had no significant influence on the content of Glu and GABA.On the 28th day,the content of Glu and GABA were kept on a higher level in model group,and decreased significantly in low-dose SalB group and high-dose SalB group compared with model group(P0.05).Conclusion SalB can significantly reduce the content of striatal Glu and GABA in rats with I/R injury as the prolonging of cerebral ischemia,and the protective effect of SalB may be related to the regulation of content of Glu and GABA.
