摘 要:目的通过链脲佐菌素(STZ)诱导建立糖尿病大鼠模型,探讨不同时间段给予抵挡汤干预对糖尿病大鼠视网膜病变基因表达的影响及其作用机制。方法高脂饲料喂养大鼠12周时尾静脉注射STZ制备2型糖尿病大鼠模型,抵挡汤早期组、抵挡汤中期组和抵挡汤晚期组分别于8周、13周和16周给予抵挡汤灌胃,辛伐他汀组和罗格列酮组大鼠于13周分别给予辛伐他汀和罗格列酮灌胃,上述各组给药均至实验24周时结束。观察抵挡汤对糖尿病大鼠体重、血糖、血胰岛素的影响,采用逆转录聚合酶链反应法(RT-PCR)检测糖尿病大鼠视网膜组织中血管内皮生长因子(VEGF)-mRNA和蛋白激酶C(PKCβ2)-mRNA表达水平的变化。结果实验第13周时,与正常对照组比较,模型组大鼠空腹血糖明显升高(P<0.01),大鼠视网膜VEGF-mRNA与PKCβ2-mRNA表达量也显著升高(P<0.01)。与模型对照组比较,抵挡汤给药干预的3组大鼠视网膜VEGF-mRNA与PKCβ2-mRNA表达有显著降低(P<0.05);与抵挡汤晚期组和抵挡汤中期组比较,抵挡汤早期组VEGF-mRNA与PKCβ2-mRNA的表达明显降低(P<0.05)。与模型对照组相比,辛伐他汀组和罗格列酮组大鼠视网膜VEGF-mRNA与PKCβ2-mRNA表达降低(P<0.05)。结论抵挡汤早期干预可延缓糖尿病大鼠视网膜病变的发展,这与抵挡汤降低大鼠视网膜VEGF和PKCβ2基因表达从而抑制炎症反应有关。Objective To investigate the influence of intervention of Didang Tong at different time points on gene expression in rats with diabetic retinopathy and the mechanism through establishing rat model of diabetes induced by streptozotocin(STZ).Methods The rat model of diabetes was established by the injection of STZ in caudal vein after high fat forage for 12 weeks.The groups with Didang Tong at early term,middle term and late term(early-term group,mid-term group and late-term group) were,respectively,given intragastrically Didang Tang on the 8th week,13th week and 16th week,and simvastatin group and rosiglitazone group were,respectively,given intragastrically simvastatin or rosiglitazone on the 13th week.After 24 weeks,the influences of Didang Tang on weight,plasma glucose and serum insulin were observed.The expression changes of retinal vascular endothelial growth factor(VEGF) mRNA and protein kinase C(PKC β2) mRNA were detected by using reverse transcription-polymerase chain reaction(RT-PCR).Results On the 13th week,the level of fasting plasma glucose(FPG) increased significantly(P0.01),and expressions of retinal VEGF mRNA and PKCβ2 mRNA increased significantly(P0.01) in model group compared with normal group.Compared with model group,the expressions of retinal VEGF mRNA and PKCβ2 mRNA decreased significantly in early-term group,mid-term group and late-term group(P0.05).Compared with late-term group and mid-term group,the expressions of retinal VEGF mRNA and PKCβ2 mRNA decreased significantly in early-term group(P0.05).Compared with model group,the expressions of retinal VEGF mRNA and PKCβ2 mRNA decreased in simvastatin group and rosiglitazone group(P0.05).Conclusion The early intervention of Didang Tang can delay the development of diabetic retinopathy in diabetes rats,which is related to that Didang Tang can reduce the gene expressions of retinal VEGF and PKCβ2 and inhibit inflammatory reaction.
