摘  要:研究了从甜茶叶中纯化甜茶苷的制备工艺。首先用沸水从甜茶叶中提取甜茶苷,提取3次,每次1 h。再利用HP-20大孔吸附树脂对甜茶苷粗提物进行预纯化,以90%乙醇为洗脱液,洗脱6倍柱体积;浓缩去除乙醇后,用D-941阴离子交换树脂脱色,水洗并干燥得到甜茶苷粗产品,纯度为68.6%。最后甜茶苷粗产品经制备色谱纯化后,得到纯度为97.9%的甜茶苷,色谱条件为流动相V甲醇∶V水=70∶30,流速10 mL/min,上样量200 mg。The preparative separation of rubusoside from the leaves of Rubus suavissimus S. Lee has been investigated. First, the leaves were extracted three times using boiling water, with an extraction time of 1 h. Second, HP-20 maeroporous resins were used in the pre-purification procedure, with 6BV of 90% ethanol in water used as the eluent. Third, the solution was evaporated to remove ethanol and loaded onto pretreated D-941 anion-exchange resin for deeoloration. Finally, preparative HPLC was used for further purification. Rubusoside with a purity of 97.9% was obtained from the 68.6% purity rubusoside extract after the preparative HPLC using 70% methanol in water as the mobile phase, a flow rate of 10 mL/min and a sample loading of 200 rag.
