摘 要:该文研究了哮喘豚鼠嗜酸性粒细胞 (EOS)在外周血及支气管肺泡灌洗液 (BALF)中的变化 ,用RT -PCR半定量测定了外周血单个核细胞 (PBMC)及支气管组织白细胞介素 5 (IL - 5 )mRNA表达量。结果显示哮喘豚鼠EOS及IL - 5mRNA表达量均较对照组及地塞米松干预组显著升高 ,PBMC中IL - 5mRNA表达量与支气管组织IL -5表达及外周血EOS计数成正相关。提示哮喘豚鼠PBMC、IL - 5mRNA表达增强 。Objective: To investigate the state and significance of IL-5mRNA expression in peripheral blood mononuclear cells (PBMC) of asthmatic guinea pigs. Methods: 31 guinea pigs were randomly allocated into 3 experimental groups, viz. The blank control group (C group n=10), the asthmatic group (A group n=11), and the dexamethasone (DXM) treatment group (D group n=10). The A and D groups were challenged by ovalbumin (OVA) to produce asthmatic models. Whereas the D group were received intraperitoneal DXM before each OVA aerosol challenge. Peripheral white blood cell and differential count were compared with that of the BALF of the experimental animals. PBMC and bronchial tissue IL-5mRNA expression level was measured semiquantitatively by reverse transcriptional polymerase chain reaction (RT-PCR). Results: The eosinophil count in blood and BALF of A group were significantly higher than the C group ( P <0.01) and D group ( P <0.005 and P <0.01 respectively). The PBMC IL-5mRNA expression level of A group (84.10+26.75%) was significantly higher than that of the C group (40.80+13.90%) and the D group (59.50+9.22%), P <0.05, P <0.001 respectively. The PBMC IL-5mRNA expression level was positively correlated with that of the bronchial tissue and blood eosinophils of the asthmatic guinea pigs, r =0.705, P <0.01; r =0.558, P <0.01. Conclusions: The PBMC IL-5mRNA expression can reflect the state of eosinophils infiltration and IL-5mRNA expression increased inflammatory pathology in the airway of asthmatic guinea pig models. Measurement of IL-5mRNA expression by PBMC may become a new clinical test that is useful in understanding the severity of asthma and the reaction to corticostoroid therapy.
