摘　　要：目的:探讨人蛋白酶体β亚基4型(PSMB4)与Bcl-2家族中的促凋亡蛋白分子Bim之间的相互作用。方法:借助PlexA-bimL技术从人胎脑文库筛选PSMB4,借助显微共聚焦技术探讨PSMB4与Bim相互作用的亚细胞定位,应用免疫共沉淀技术探讨PSMB4与Bim的相互作用。结果:PSMB4与Bim存在于同一细胞中,PSMB4和Bim在空间上存在相互作用。结论:PSMB4和Bim有可能存在空间位置上的共定位;PSMB4在细胞内能够与Bim发生相互作用。Objective:To find out the interaction of PSMB4 and BIM(a member of Bcl-2 family protein). Methods:Screen out PSMB4 by using plexA-bimL;discuss the interaction of PSMB4 and BIM through the coimmunoprecipitation;with the help of confocal microscope technology,mouse out the subcellular localization of the interaction.Results:The consequence of coimmu-noprecipitation: in HEK293 cells,PSMB4 which.is of exogenous expression can' interact on GFP-tagged BimL,especially in those apoptotic cells which are over expressed in protein Bim.Conclusions:PSMB4 and BimL may have co-localization.The interaction of PSMB4 and BimL could happen in cells.
