摘  要:目的:观察聚焦超声辐照对神经生长因子(NGF)和P物质(SP)在豚鼠慢性湿疹模型皮肤表达变化的影响.方法:将40只豚鼠随机分成2组,正常组和建模组,每组20只.建模组用2,4-二硝基氯苯制作慢性湿疹动物模型后,自身空白对照,用功率2W聚焦超声辐照一侧,作为辐照组,另一侧空照,作为对照组.超声辐照前,正常组和建模组取皮肤活检,超声辐照后14天,辐照组和对照组取皮肤活检.用SP免疫组化法观察NGF和SP的表达变化.结果:正常组NGF和SP主要表达于表皮中上层,阳性染色浅黄色至棕黄色.建模组,皮损中NGF和SP表达于表皮全层,阳性染色呈棕黄色至棕褐色,明显高于正常组(P〈0.001).超声辐照后14天,辐照组NGF和SP表皮角质形成细胞阳性染色变淡,而对照组无明显改变(P〈0.001).NGF和SP二者阳性表达呈正相关(P〈0.05).结论:在慢性湿疹豚鼠模型中,表皮角质形成细胞NGF和SP表达增强.聚焦超声辐照后表皮角质形成细胞NGF和SP的分泌和表达明显降低.Objective:To observe the influence of nerve growth factor and substance P after focused ultrasound irradiation on guinea pig model of chronic eczema. Methods: 40 guinea pigs were divided into 2 equal groups. 20 guinea pigs were prepared by using 2,4--dlnitrochlorobenzene repeated- ly and treated one shaved site but not the other shaved site by 2W focused ultrasound irradiation. The other 20 guinea pigs were normal control group. Samples were taken before ultrasound irradiation and on 14 days after irradiation. The immunohistochemical methold was used to detect the,. changes of NGF and SP protein in the skin of guinea pigs. Result:Both NGF and SP protein in the epidermis were higher distinctly in the model group than in the normal group(P〈0. 001). On 14 days after ultrasound irradiation the significant difference between the treatment group and the control group was also observed in the expression of NGF and SP protein(P〈0. 001). There was a positive correlated signification between NGF and SP(P〈 0. 05). Conclusion:NGF and SP play an important role in the pathogenesis of chronic eczema. Controlling the secretion and expression of NGF and SP may be one of the mechanisms of focused ultrasound treating the chronic eczema.
