摘　　要：目的建立RP-HPLC法同时测定金银花药材中酚酸类(绿原酸、咖啡酸)、环烯醚萜类(马钱苷)和黄酮类(芦丁、金丝桃苷、木犀草苷、槲皮素)7种主要成分,并测定不同变异类型花蕾中各成分,为金银花药材的多指标、多成分质量控制体系建立奠定基础。方法采用Aglient TC-C18色谱柱(4.6 mm×250 mm,5μm),流动相A为0.4%磷酸水溶液,流动相B为乙腈,梯度洗脱程序为0～18 min,A为87%;18～30 min,A为85%;30～43 min,A为85%～80%;43～48 min,A为80%;48～50 min,A为80%～70%;50～55 min,A为70%;55～65 min,A为70%～50%;检测波长为0～16 min,240 nm;16～40 min,323 nm;40～65 min,355 nm;体积流量1.0 mL/min,柱温为30℃。结果 7种成分均达到了基线分离,且各成分在线性范围内线性关系良好;7种成分的加样回收率变化范围为99.56%～102.12%。结论该方法准确、简便、可行,可以用于不同变异类型金银花花蕾7种活性成分的测定。AIM To set up a new HPLC method for simultaneously determining seven major components of different variants of Lonicerae japonicae Flos,belonging to phenolic acids（chlorogenic acid and caffeic acid）,iridoids（loganin） and flavonoids（rutin,hyperin,galuteolin and quercetin）.METHODS The Aglient TC-C18 column（4.6 mm×250 mm,5 μm） with the column temperature of 30 ℃ was used in RP-HPLC.The mobile phase was composed of 0.4% phosphoric（A） and acetonitrile（B） using a gradient elution of 87%A within 0-18 min,85%A within 18-30 min,85%-80%A within 30-43 min,80%A within 43-48 min,80%-70%A within 48-50 min,70%A within 50-55 min and 70%-50%A within 55-65 min.The flow rate was 1.0 mL/min and detection wavelength was set as follows： 240 nm within 0-16 min,323 nm within 16-40 min and 355 nm within 40-65 min.RESULTS Seven compositions were separated well,all calibration curves showed the good linear regression within test ranges and their recoveries were found to range from 99.56%-102.12%.CONCLUSION The method is accurate,reliable and convenient.It can be applied to evaluating the quality of Lonicerae japonicae Flos from diffenent variants.
