摘  要:目的 探讨慢性铅暴露对小鼠脑海马神经元蛋白激酶B表达的影响。方法 5~6周龄小鼠交配后,通过饮水饲以终浓度(0,2.4,4.8,9.6mmol·L^-1)醋酸铅制备动物模型。用Western blot法观察各组小鼠脑海马神经元总量PKB(t-PKB)及磷酸化PKB(p-PKB)的表达变化。结果 染铅组小鼠血铅和脑铅浓度明显升高,与对照组比较有统计学意义(p<0.01)。染铅组脑海马神经元p-PKB表达水平比对照组明显降低,并与血铅浓度有效应关系,呈负相关(r=-0.84,p<0.01)。铅对小鼠大脑海马神经元细胞t-PKB的表达水平没有影响。结论 慢性铅暴露导致小鼠脑海马神经元中p-PKB表达的改变可能与小鼠学习记忆功能减退有关。Objective To explore the effect of chronic lead exposure on expression of protein kinase B(PKB) in hippocampus of mice. Methods Chronic acetic lead exposure was applied in mice (0, 2.4, 4.8, 9.6mmol·L^-1) for 6 weeks. At the end of 6 weeks, the expression of total PKB (t-PKB) and phosphorylated PKB (p-PKB) was determined by Western blots. Results The lead concentrations in blood and brain were higher in lead exposure group than in control group (p〈 0.01). The expression level of p-PKB in hippocampus was decreased in lead exposure group than in normal control with negative correlation with concentrations of blood lead ( r=-0.84, P 〈0.01 ) , but t-PKB protein levels did not change under the same experimental conditions. Conclusion The decreased expression level of p-PKB in hippocampus neuron might be related to the damage of learning and memory induced by lead in mice.
