摘 要:目的建立TNF-α抑制剂的高通量药物筛选模型。方法用对TNF-α敏感的L929细胞株通过MTF法建立TNF-α抑制剂的筛选模型;筛选了样品并对筛得的活性样品进行进一步的评价。结果建直TNF-α抑制剂L929细胞株筛选模型;初筛样品数:870个,复筛样品数:47个,活性药物:8个;对其中的鹿衔草做了淋巴细胞转化抑制实验。结论TNF-α抑制剂的L929细胞株筛选模型经济稳定可以用于高通量药物筛选;鹿衔草可能具有抑制TNF-α作用和一定的免疫调节作用。Objective To establish a high-throughput drug screening model of TNF-α inhibitors. Methods A screening model for inhibitors of TNF-α was established by adopting TNF-α-sensitive L929 cell line with MTT assay. The active samples were further analyzed and evaluated. Results TNF-α inhibitor L929 cell line screening model was utilized for primarily screening of 870 samples, re-screened 47 samles and 8 active drugs, as well as the experiment of Chinese Pyrola herb transformation inhibition assay. Conclusion TNF-α inhibitors L929 cell line screening model with its feature of the low cost and being stable is recommended for high-throughput drug screening. Chinese Pyrola herb may inhibit TNF-α role and carry certain immunomodulatory effect.
