摘 要:目的了解我国不同鼠疫自然疫源地鼠疫耶尔森菌的单核苷酸多态性(SNP)特征。方法利用Mummer程序对已知全基因组序列的9株鼠疫菌进行全基因组比对,选择其中的13个基因片段(19个SNP位点)在国内133株鼠疫菌中进行PCR扩增、测序,并对序列进行UPGMA聚类分析。结果全基因组比对共筛查出3780处序列差异位点。通过对不同SNP位点组合的聚类分析发现我国鼠疫菌具有明显的地理区域性和生态型聚集性特征。结论SNP作为一种比较稳定的遗传标记,可以较好地反映我国不同鼠疫自然疫源地耶尔森鼠疫菌的基因组特征。Objective To study the single nucleotide polymorphisms (SNP) characteristics of Yersinia pestis strains from different natural foci in China. Methods Genome-wide comparison was done to find SNP sites by the Mummer program among 9 Yersinia pestis genome which was downloaded from NCBI. Then 13 genic fragments including 19 SNP sites were amplified by PCR and sequenced in 133 Yersinia pestis strains, and the results were cluster analyzed with the BioNumerics software. Results . Three thousand seven hundred and eighty sequence variation sites were found by genome-wide comparison. Using the different combinations of SNP sites, UPGMA cluster analysis revealed obvious geographic regional and eco-aggregation characteristics of Yersinia pestis strains isolated from China. Conclusions As relatively stable genetic markers, SNP can better reflect the genome characteristics of Yersinia pestis in different plague natural foci of China.
