摘 要:在pH=7.40的缓冲条件下,采用荧光光谱法和紫外-可见分光光度法研究了橙黄与牛血清白蛋白(BSA)的相互作用,研究发现橙黄对BSA有较强的荧光猝灭作用,用Stern-Volmer和Lineweaver-Burk方程分别处理实验数据,得到了猝灭常数Ksv、Kq、热力学参数(ΔHθ,ΔSθ,ΔGθ)和结合位点数等,证明二者主要靠范德华作用和氢键作用,同时用同步荧光光谱法探讨了橙黄对BSA构象的影响。The interaction between orange Ⅳ and bovine serum albumin(BSA) was investigated by fluorescence spectrometry and UV-Vis spectrometry under the buffer condition with pH of 7.40.The quenching mechanism was analyzed referring to orange Ⅳ against the fluorescence of BSA.The experimental data was deal with by Stem-Volmer equation and Lineweaver-Burk equation,then the quenching constants Ksv,Kq,the thermodynamic parameters(ΔH^θ,ΔS^θ,ΔG^θ) and the number of binding sites were obtained,and the main forces between orange Ⅳand BSA were Van der Waals and hydrogen bind,meanwhile,the influence of orange Ⅳ on BSA was investigated by synchronous fluorescence spectrometry.
