摘  要:目的:观察大鼠骨髓间充质干细胞在子宫内膜组织的迁移分化情况。方法:第2代雄性大鼠骨髓间充质干细胞行PKH26标记,实验组5只雌性大鼠尾静脉注射1×107/mlPKH26标记BMSCs 1 ml,对照组5只雌性大鼠尾静脉注射生理盐水1 ml。6周后切下子宫制成冰冻切片,荧光显微镜下观察标记细胞在子宫内膜组织的分布情况,并进行免疫荧光染色检测标记细胞角蛋白的表达。结果:实验组大鼠子宫内膜组织均可见标记细胞散在分布于子宫内膜组织中,腺上皮中少数标记细胞角蛋白染色阳性。对照组子宫内膜均未见标记细胞。结论:BMSCs可以迁移至子宫内膜,可以向腺上皮细胞分化,参与子宫内膜的增生。Objective: To observe the migration and differentiation of rat mesenchymal stem cells in endometrial tissue. Methods: The mcsenchymal stem cells of the second generation male rats were labeled by PKH26. Five female rats in experimental group were treated with injection of 1 × 107 ml PKH 26 into caudal vein to label mesenchymal stem ceils, while five female rats in control group were treated with injection of 1 ml normal saline into caudal vein. Uterine frozen sections were prepared after six weeks, and then the distribution of mesenchy- real stem cells in endometrial tissue was observed under fluorescence microscope, the expression of keratin in mesenchymal stem cells was detected by immunofluorescent staining. Results: The mesenchymal stem cells labeled by PKH 26 distributed in endometrial tissue of rats diffusely in experimental group, the expression of keratin was positive in a few mesenchymal stem cells labeled by PKH 26 in glandular epi- thelium. The mesenchymal stem cells labeled by PKH 26 didnt express in endometrium of rats in control group. Conclusion : The mesenehy- real stem cells can migrant to endometrinm, differentiate into glandular epithelial cells, and participate in proliferation of endometrial cells.
