兔IL-6真核表达质粒的构建及其分子佐剂效应的初步研究

摘　　要：为了研究兔IL-6真核表达质粒(pcDNA-IL-6)对家兔免疫应答的影响,本文构建了真核表达质粒pcDNA-IL-6,并将其与pcDNA-VP60、兔瘟组织灭活苗分别共同免疫家兔,用血凝抑制试验检测家兔RHDV特异性抗体水平。结果表明:在免疫后的7 d到70 d,无论是核酸疫苗pcDNA-VP60或是兔瘟组织灭活苗,与真核重组质粒pcDNA-IL-6共注射免疫组抗体水平均高于未注射pcDNA-IL-6质粒免疫组,差异均显著P<0.05或极显著P<0.01。真核重组质粒pcDNA-IL-6对重组质粒pcDNA-VP60和组织灭活苗的免疫均具有免疫增强作用。The aim of the study was to investigate how the pcDNA-IL-6 effects immune response in rabbits.in this research,the pcDNA-IL-6 were constructed and immuned rabbits with pcDNA-VP60 and RHD tissue inactivity.Hemagglutination test was used to detect antibodies against RHDV.after vacination,from 7th d to 70th d,the immune responses reduced by pcDNA-VP60＋ pcDNA-IL6 and RHD inactivitied tissue vacine＋ pcDNA-IL6 significant stronger than by pcDNA-VP60 and RHD inactivitied tissue vacine（P0.05 or P 0.01）.Research results indicated that：the immune effect of the pcDNA-VP60 and RHD inactivited tissue vacine can be enhanced by the pcDNA-IL-6;
