摘 要:采用3×3双因子设计,研究发根农杆菌菌株和烟草品种对烟草毛状根诱导率的影响。结果表明:各菌株在不同烟草品种上均诱导出毛状根,经PCR技术检测,确定了发根农杆菌Ri质粒的rolB基因已整合到烟草基因组中。不同烟草品种和发根农杆菌菌株均显著影响毛状根的诱导率;Sumxun毛状根平均诱导率最高,达到了53.4%,其适宜菌株为A4和ATCC15834;K326为34.2%,其适宜菌株为R1000;云烟85为25.6%,其适宜菌株为ATCC15834。Different Agrobacterium rhizogenes strains and tobacco cultivars on the transformation of hairy root were investigated by 3 × 3 factor design experiments. The PCR technique was employed to detect hairy root. The results showed that rolB gene in Ri plasmid of A. rhizogenes strains were integrated into the genome of cell of tobacco and the hairy roots were induced successfully. Different rhizogenes strains and tobacco cultivars influenced the transformation rate of hairy root. The average transformation rate of Sumxun ( up to 53.4% ) was the highest among the tested cultivars, whith suitable bacterial strains A4 and ATCC15834. The transfor- mation rate with strain R1000 of cultivar K326 was 34.2% , and that of cultivarYunyan85 was 25.6% with strain ATCC15834. The results might provide an experimental basement for the induction of tobacco hairy roots via transformation.
