摘 要:目的探讨脂肪源性干细胞(ADSCs)对大鼠阴道黏膜下生物补片生物相容性的影响。方法 2010年4月至2011年2月在广州医学院第一附属医院,获取大鼠腹股沟处脂肪组织分离培养ADSCs,选择第3代细胞与脱细胞猪心包膜(APP)、脱细胞牛心包膜(ABP)两种生物补片构建新型组织工程补片,选取体外培养1、7d的新型组织工程补片进行扫描电镜。根据大鼠阴道黏膜下植入生物补片的不同类型,将20只去势SD雌性大鼠随机分为5组:APP组,ABP组,APP+ADSCs组,ABP+ADSCs组,对照组。术后30d植入部位取材,HE染色观察组织的炎症反应程度,巨噬细胞及异物巨细胞计数。结果 ADSCs可以在两种生物补片上黏附及生长。与ABP组比较,APP组局部的炎症反应及异物反应较重,补片结构破坏明显。加载ADSCs的两种生物补片植入局部组织的炎症反应及异物反应明显低于未加载ADSCs的生物补片。结论 ABP较APP具有更好的生物相容性。ADSCs可以调节生物补片植入部位的炎症反应及异物反应,可能对生物补片的降解有缓解作用。Objective To explore the effects of adipose-derived stem cells (ADSCs) on biocompatibility of biological patch implanted in the rats vaginal submucosa. Methods In the First Affiliate Hospital of Guangzhou Medical University between April 2010 and February 2011, ADSCs were separated from rat inguinal fat pads and cultured. The third genera- tion cells were used for constructing a new tissue engineering scaffold with acellular porcine pericardium (ABP) and acel- lular bovine pericardium (ABP). The two new tissue engineering scaffolds which were cultured one day and seven days in vitro were ready for scanning electron microscopy. Twenty Castration SD female rats were randomly divided into five groups by the biological patch implanted in the rats vaginal submucosa: APP group, ABP group, APP + ADSCs group, ABP + ADSCs group, blank control group. Specimens were collected in the place where the scaffold was placed, after 30 days of operation. Specimens were used for HE stain. To evaluate the degree of inflammation and count the number of macrophages and foreign body giant cells by HE stain. Results ADSCs can attach to the AP and grow well. Compared APP group with ABP group, the APP group has high degree of inflammatory reaction and foreign body reaction, and worse patch structure. Compared with two biological patch groups, the scaffold with ADSCs has lower degree of inflammatory re- action and foreign body reaction. Conclusion Compared with APP, ABP has better biocompatibility. ADSCs can adjust the inflammatory reaction and foreign body reaction induced by biological patch, may be can slow down the absorption of biological patch.
