摘 要:目的 通过对脂质过氧化物和心肌高能磷酸化合物的测定,评价同种异位鼠心移植模型做为缺血/再灌注损伤研究模型的可行性。方法 取SD大鼠随机分为两组(n=10),组Ⅰ:为缺血心肌组,供心缺血40分钟。组Ⅱ:为缺血再灌注心肌组,供心主动脉及肺动脉分别与受体腹主动脉和下腔静脉吻合,取复跳后40分钟的供心。结果 组Ⅱ的MDA含量高于组Ⅰ而SOD活性低于组Ⅰ(P<0.05),组Ⅰ的ATP、CP含量高于组Ⅱ(P<0.05)。结论 同种异位鼠心移植模型与临床体外循环手术过程相接近,能够真实反映心肌缺血再灌注损伤的病理生理过程,可做为心肌保护的研究模型,而且该模型具有人力需求少,设备简单,模型制做方便,节约经费等优点。Objective To study whether the heterotopic allografted rat heart model can be used as animal experiment model in ischemia/ reperfusion injury research, the model is evaluated by studying the change of peroxidation products and high-energy phosphate. Method Sprague Dowly rats are divided into two groups randomly. Group Ⅰ :ischemic hearts(n= 10), after being infused with cardioplegia, donor hearts are reserved in hypothermia (4℃) for 40 minutes. Group Ⅱ :ischemic/reperfusion hearts(n=10), donor hearts are cut off after having being transplanted for 40 minutes.Result The data show that the malondialdehyde(MDA) of group Ⅱ is higher than group I and the activity of superoxide dismutase (SOD) of group Ⅱ is lower than group Ⅰ (P< 0.05). Both adenosine triphosphate(ATP) and creatine phosphate (CP) of group Ⅱ is lower than group Ⅰ (P< 0.05) .Conclusion The heterotopic allografted rat heart model can be used as a animal experiment model in cardiac protection research for it reflect the pathophysiological process of ischemic/reperfusion injury and is similar to extracorpreal operation. It is also need less manpower and equipment, in addition, it is economical and is easy to be mastered.
