Effects of Early Liquid Resuscitation Combined with Hydrogen Inhalation on LPS-induced Gut Injury in Septic Shock Rats

摘  要:目的研究早期液体复苏联合2%氢气吸入的治疗方案对脂多糖诱导脓毒性休克大鼠小肠的作用。方法 60只Wistar大鼠随机分成4组:正常对照组(A组)、休克对照组(B组)、早期液体复苏组(C组)和早期液体复苏联合2%氢气吸入组(D组),B、C、D组建立脓毒性休克模型,C、D 2组给予相同的液体复苏方案,D组吸入气体为2%氢气空气混合气,其余组为空气,2h后观察肺组织HE染色并进行病理学评分,检测小肠中丙二醛、髓过氧化物酶及血清中二胺氧化酶表达水平。结果单纯液体复苏使肠壁水肿加重,小肠Chiu评分升高,联合2%氢气吸入后肠壁水肿减轻,小肠Chiu评分降低(P<0.05);单纯液体复苏使小肠氧化损伤加重,二胺氧化酶表达增加,联合2%氢气吸入后上述损伤明显减轻(P<0.05)。结论早期液体复苏联合2%氢气吸入明显改善脓毒性休克大鼠小肠功能。Objective To study the efficacy of fluid resuscitation combined with 2% hydrogen inhalation on LPS-induced gut injury in septic shock rat.Methods A total of 60 male Wistar rats were randomized to 4 groups:the normal control group(group A),septic shock control group(group B),fluid resuscitation group(group C),fluid resuscitation with hydrogen inhalation group(group D).The same liquid resuscitation was applied in group C and D.2% hydrogen was administrated in group D and air was used in other groups.Pathological changes of the guts were observed by light microscope.The levels of MDA,MPO and DAO were detected.Results HE staining indicated that gut edema was alleviated in group D compared with group C.Levels of MPO,MDA and DAO were decreased in group D compared with group C(P 〈 0.05).Conclusion The present study demonstrated that fluid resuscitation with 2% hydrogen inhalation had a protective effect on gut injuries induced by LPS.
