摘 要:目的探讨手术创伤对老年鼠和成年鼠肝部分切除术后空间学习记忆能力的影响。方法成年和老年SD大鼠随机分为对照组、麻醉组、手术组,对照组SD大鼠不做特殊处理。麻醉组实验动物单纯接受1.5%~2%异氟醚麻醉,行气管插管和机械通气。手术组SD大鼠行肝部分切除术。Morris水迷宫用于检测实验鼠的空间学习记忆能力。结果成年鼠的空间记忆学习能力到达平台时间和游行距离在术后1 d(P〈0.001)受到破坏,在术后3 d恢复(P=0.848,P=0.973)。老年鼠的空间记忆学习能力在术后1 d(P〈0.001)受到破坏,并且持续到术后3 d(P=0.002,P=0.001),在术后7 d恢复。结论手术创伤而不是麻醉因素导致了术后认知功能障碍,高龄加剧了认知功能的破坏。Objective To investigate the effects of partial hepatectomy on spatial learning and memory in adult and aged rats.Methods Both adult(3~6 months old)and aged(20~24 months old)Sprague-Dawley male rats were randomly divided into following groups:control(n =12 for each group),anesthesia alone(n =36 for each group),and operation group(n =36 for each group).Control animals didn′t received special treatment except sterile saline.The anesthesia alone group received 1.5% to 2% isoflurane with intubation and mechanical ventilation.The operation group underwent partial hepatectomy under general anesthesia.Cognitive function was assessed in a reversal-learning version of the Morris water maze(MWM)before and after surgery.Results Surgical trauma resulted in impairments in distance and latency only on postoperative day 1(P 〈 0.001,respectively)in adult rats.Aged rats exhibited impairments on day 1(P 〈 0.001)that persisted until postoperative day 3(P =0.002 and P =0.001,respectively).Conclusion These findings suggest that surgical trauma,rather than anesthesia,resulted in cognitive function impairment which would be exacerbated by aging.
