摘 要:目的研究不同程度低氧对体外培养人胃粘膜上皮细胞株GES-1中MICA基因表达的影响。方法建立GES-1细胞体外低氧培养模型。采用RT-PCR和免疫细胞化学技术分别检测模型MICA基因和蛋白的表达变化。结果在缺氧环境下培养0、48和72 h后MICA蛋白表达(57.67±2.30、96.14±1.75、259.59±3.51)及mRNA/β-actin比值(0.22±0.03、0.37±0.02、0.68±0.03)都随缺氧时间不同表达量存在差别(P<0.05),且表达量随缺氧时间延长而升高,直线相关分析显示二者表达呈显著正相关。结论低氧可以刺激胃上皮细胞株GES-1中MICA基因的表达量显著增加。Objective To investigate the influence of different degrees of hypoxia on Major Histocompatibility Complex Class I-related Chain A(MICA)expression in Gastric Epithelial cell Strain-1(GES-1).Methods Hypoxia culture model in vitro was established to test the expression of MICA in GES-1 cell strain.After cultured in hypoxia circumstance for a given period of time(0hr,48hrs,72hrs),MICA expression was tested in mRNA level and protein level by RT-PCR and immunocytochemistry analysis.Results MICA expressions increased after 0 hr,48 hrs,72 hrs in hypoxia(P〈0.05).MICA protein expressions(57.67±2.30,96.14±1.75,259.59±3.51,respectively)increased equally with mRNA(0.22±0.03,0.37±0.02,0.68±0.03,respectively)confirmed by immunocytochemistry and RT-PCR and showed positive correlation with linear correlation analysis.Conclusion MICA gene and protein can be induced remarkably by hypoxia in GES-1 strain.
