摘  要:以异源胞质洋葱不育系及其保持系为材料,克隆洋葱atp6基因,并对其在DNA、蛋白质二级结构和花发育各时期表达量等方面进行分析比较。结果表明:获得的atp6基因全长均为1414bp,与保持系进行序列比对发现,不育系开放阅读框第92碱基处由C突变为A,该突变导致编码氨基酸由酪氨酸突变为丝氨酸;二者编码的氨基酸在二级结构组成及蛋白质残基溶剂可接触性上也存在不同程度的差异;其表达量在小花蕾和大花蕾时差异不大,中花蕾时不育系却明显高于保持系,表达上调。Cytoplasmic male sterility(CMS) plays an important role in genetic breeding research of higher plant,and has been the hot spot of the genetic breeding of plant and the genetic theory research.CMS line and its maintainer line of onion were used as experimental materials.Compared differences from atp6 gene of CMS line and maintainer line,the Real-time PCR was used to analyse its different expression in florescence.The results showed that the length of the two sequence were all 1414bp,compared with the maintainer line,the first 92 basic group of the atp6 gene open reading frame of CMS line mutated from C to A.This mutations lead tyrosine to serine.The amino acid different at composition of secondary structure and solvent-assisted composition.The level of expression in the small and big bud had little difference,but in the medium-term the male sterile line was significantly higher than that of the maintainer line.
